Purpose: This study will examine the impact of circadian misalignment (sleeping during the day and being awake and eating at night as is common in shift-workers) on cognition and metabolic physiology in adults. This research will have important implications for people who do not get enough sleep or sleep at the wrong time of day and are at a high risk for developing diabetes and obesity.

This sleep study is conducted by the Sleep and Metabolism Laboratory (SAM Lab) at Colorado State University (CSU).

Study Requirements: Healthy Men & Women Age 18-35

Visit 1: Informed Consent & Health, Sleep, and Psychological questionnaires.

Visit 2: Medical screening tests including a physical exam, blood work, an ECG, a DEXA scan to measure body composition, and a resting metabolic rate assessment (which measures calorie consumption).

You are not financially compensated for the screening procedures, but we can share them with you if you wish. All results of the screening procedures will only be used to determine eligibility for the study.

Visit 3/6: Equipment/Watch Pickup and start 8-hour sleep/wake schedule one week before in-lab stay.

Visit 4/7: Diet Pickup that you eat 3 days before your in-lab stay.

Visit 5/8: In-lab Study visit includes living in the SAM Lab at CSU for 5 days. Meals will be provided.

Day 1- Arrive to the lab in the afternoon on Day 1 and receive a normal night of sleep. In the morning, remain in dim light and in bed and have blood sampled every hour.

Day 2- Trained professional performs muscle biopsy to see how your muscle functions. An oral glucose tolerance test will also be done which involves drinking a sugary drink to see how your body responds to glucose.

Day 3&4- Follow a “shiftwork protocol” which means sleeping during the day and being awake at night.

Day 4&5- Repeat blood sampling, muscle biopsy and glucose tolerance test.

During your stay, you'll also complete computer tasks to measure your ability to think, complete strength & balance tests, and wear EEG to measure your brain activity. Roughly 50% of your time in the laboratory will be free time. After the testing is complete, you'll receive a 12-hour recovery sleep opportunity.

One month later, you will come back to the lab for your second in-lab stay and complete vists 6/7/8.

The only difference between your first and second in-lab visits is that in one visit you will eat food during the nighttime hours during the “shift work” days and in the other visit, you will only eat during daytime hours.

If you are eligible to participate in this study, you will be financially compensated for your involvement after the screening procedures. The total compensation can be up to $2,650.

This sleep study is conducted by Colorado State University, the Sleep and Chronobiology Laboratory at the University of Colorado at Boulder, and the University of Colorado Denver.

Study Requirements: Healthy Men & Women Age 18-35

Purpose: This study is about the effect of insufficient sleep on physiology in people. Sleeping too little and being awake at the wrong time can impact your ability to think, proteins and hormones in your blood, how your muscle and fat cells work and many other things. This work will have important implications for people who do not get enough sleep and are at a high risk for developing diabetes and obesity.

Visit 1: Consent & Screening Questionnaires

Visit 2: Medical Screening including a physical exam, blood work, an electrocardiogram (a measurement of heart activity), and a resting metabolic rate assessment (oxygen consumption).

Although you are not financially compensated for the screening procedures, we do cover the expense of the
screening tests. All of the results of the screening procedures are confidential, and will only be used by the study staff to determine eligibility for the study.

Visit 3 & 4 (after determined eligible): Equipment Pickup/Exercise Test & MRI
We ask you to wear a special watch that records activity levels for one week prior to your stay in the study. We also ask you to keep a 9-hour sleep schedule, a sleep/wake log, and text us when you go to bed and wake up each day. One day during the week before check-in, we will have you come in for an exercise test on a bike, a MRI of your calf muscle, and provided meals that you will eat for 3 days before checking in.

Visit 5: Laboratory study stay
During your laboratory study visit, you will live in the Clinical and Translational Research Center at the University of Colorado Denver in Aurora for 7 days and 7 nights. You will come to the laboratory approximately 5 hours before bedtime on Day 1. During the study your sleep schedule will be changed to simulate the sleep loss that 30% of Americans typically experience. You will be given 5 hour sleep opportunities for 5 nights. We will take blood and saliva samples to test for circadian and metabolic markers. We will have you do some computer tasks periodically and we will also test how your body responds to sugar. The day before you check-out, you will repeat the exercise test and MRI on your calf muscle. At the end of the study visit you will be given a 12 hour sleep opportunity for recovery.

If you are eligible to participate in this study, you will be financially compensated for your involvement after the screening procedures. You will be compensated for the week of activity monitoring, each day in the laboratory, the exercise tests, the MRIs, and for transportation to and from the laboratory for each study visit. The total compensation can be up to $1,475.

This sleep study is conducted by the Sleep and Metabolism Laboratory at Colorado State University.

Study Requirements: Healthy Men & Women Age 35-55 with sleep quality complaints

This research study is aimed to answer whether or not oral cannabinoid (CBD) treatment has therapeutic effects on sleep quality. Adults aged 35-55 with reported sleep problems will be studied.

Activity in this research study includes three visits,

(1) a consent visit with health questionnaires given to determine eligibility,

(2) an equipment and treatment dose (powder packet) pickup (You will be randomized to a CBD dose that will be between 0-40mg.), and finally

(3) an equipment and remaining treatment dose drop-off at completion of the study.

The study will take approximately 3-4 weeks to complete. There are some risks to participating in this study, like nausea, fatigue and irritability. We hope that this research will benefit the sleep quality of people with sleep problems and ultimately reduce the risks of health problems associated with insufficient sleep.
